The Eagles selected him in the first round of the 2012 NFL Draft after he played college football for Mississippi State. Cox won the Super Bowl and was chosen six times for the Pro Bowl. He participated in the high jump and the relays for the Yazoo High track and field team. He finished eighth in the high jump at the 2009 Division 3-4A Meet with a leap of 1.74 meters. His personal-best times in relays were 44.32 seconds for the 4 100-meter relay and 1:32.63 minutes for the 4 200-meter relay.  In addition, he benched 300 pounds and ran 40 yards in 4.47 seconds.

In the first round of the 2012 NFL Draft, the Philadelphia Eagles selected Cox 12th overall after moving up from the 15th pick. Since Michael Haddix in 1983, he is the most highly selected defensive lineman at Mississippi State, and since Jimmy Webb in 1975, he is also the most highly selected Bulldog overall. On June 18, 2012, Cox agreed to a four-year contract. After punching Detroit Lions players on October 14, 2012, Cox was dismissed from the game and fined $21,000. Cox played 15 games in his first season of 2012 and recorded 39 tackles, 5.5 sacks, 4 passes defended, and 1 forced fumble.
