In Sarasota, Florida, James attended Riverview High School. He participated in track and earned a letter in addition to playing football, winning numerous state championships. In December 2013, James made a commitment to play football for the Georgia Southern Eagles, but he later changed his mind due to a coaching change. In January 2014, he made a decision to play football for the Middle Tennessee Blue Raiders, and in February that year, he signed.

James chose to redshirt rather than play as a true freshman in 2014. In 2015, as a redshirt freshman, James appeared in all 13 games for Middle Tennessee, catching 108 passes for 1,346 yards and eight touchdowns. On 12 attempts, he also rushed for 146 yards and one touchdown. He was named to the C-USA All-Conference First Team in 2015 and the C-USA All-Freshman Team in 2015.

In 2016, James appeared in all 13 games once more, catching 105 passes for 1,625 yards and 12 touchdowns while also rushing for 339 yards and four touchdowns on 38 attempts. He was named to the C-USA All-Conference First Team in 2016.

In 2017, James appeared in five games as a redshirt junior, catching 31 passes for 290 yards and three touchdowns. He broke his collarbone against Marshall, however, and was ruled out for the season. Despite limited playing time in 2017, James established himself as the Middle Tennessee career leader in receiving yards (3,261) and receiving touchdowns (23). James declared for the 2018 NFL Draft following the season.
