Hankin was born in New York City on December 7, 1937, and grew up in the Far Rockaway neighborhood of Queens in a Jewish family. Carl Gottlieb met him at Far Rockaway High School and Syracuse University.

Hankin was a founding member of the Second City, studying improvisation with Viola Spolin and Paul Sills before moving to San Francisco in 1963 to co-found The Committee. In 1969, he played Pt. Romero in Viva Max!, his first notable supporting role.

He appeared in Escape from Alcatraz in 1979, Running Scared in 1986, and Billy Madison in 1995. His small roles included Joe in Breaking Bad and El Camino, Sergeant Larry Balzak in Home Alone, Doobie in Planes, Trains, and Automobiles, and Mr. Heckles in Friends.

Hankin played Tom Pepper on Seinfeld and Kramer on Jerry, a pilot within a show. He played a homeless man in Malcolm in the Middle Season 5. In Breaking Bad seasons three and five, he played junkyard owner Old Joe alongside Bryan Cranston.
