On February 8, 1939, José Sison was born in Cabugao, Ilocos Sur, to a well-known landowner family with mestizo (Spanish, Mexican, Malay) and Chinese ancestry.
Sison earned a Bachelor of Arts in English literature with honors from the University of the Philippines in 1959.
After studying Indonesian there, he returned to the Philippines and went on to become a university professor of literature, Rizal Studies, and Political Science.

At the end of the 19th century, Don Leandro Serrano, his great-grandfather, was the largest landowner in northern Luzon.
Teófilo Sison, who was his uncle, was the first Defense Secretary in the Commonwealth administration and the governor of Pangasinan.

His wife is Julie de Lima. They were both products of UP Diliman. They were even in the same study group.
